Reconstitution Basics for Laboratory Peptides
Reconstitution, dissolving a lyophilized peptide into a liquid solution, is a routine step in most research workflows, but doing it carelessly can waste an otherwise high-purity sample. A few basic principles keep the process consistent.
Choosing a diluent
The right diluent depends on the specific peptide and the intended experimental use, and researchers should refer to the product-specific documentation or established literature for their compound. Common choices include bacteriostatic water, sterile water, or dilute acetic acid for peptides with poor water solubility. Whatever diluent is used, it should be added carefully rather than in one motion.
Technique matters
Add the diluent slowly, aiming it down the inside wall of the vial rather than directly onto the lyophilized powder. Introducing liquid too forcefully can denature the peptide before it’s even fully in solution. Let the vial sit briefly, then gently swirl; avoid vigorous shaking, which can introduce air and stress the peptide structure.

After reconstitution
